چکیده
Mass spectrometry combined with the use of isotope-labeled standards is one favored methods for quantitative measurement beta-amyloid concentrations in biological media. This article devoted to prevention typical systematic errors such measurements arising from neglect possibility transformation into denatured form, which gives mass spectra signals three times more intensity than native form. The degree this denaturation was determined by ability form set up a complex α-2-macroglobulin. lacks point. It shown that can occur case heat treatment an acidic environment or when DMSO used as solvent. Measurement error occurs standard and analyte are different forms. There suggested recommendations overcome analysis these compounds forcing mixture before analysis.

P135: The Role of Amyloid Beta-Peptides and Tau Protein in Alzheimer\'s Disease
Alzheimer's desease is the most common age-related neurodegenerative disorder, and cognitive problems such as defects in learning and memory are of its symptoms. Among the factors involved in the pathogenesis of the disease are biochemical disorders in protein production, oxidative stress, decreased acetylcholine secretion and inflammation of the brain tissue. Extra-neuronal accumulation ...
متن کاملMembrane disordering effects of beta-amyloid peptides.
The interaction of Abeta with synaptosomal plasma membranes decreases membrane fluidity. Using model membrane/liposome systems the interaction of Abeta with specific lipids (e.g. phospholipids, gangliosides, cholesterol) has been defined. The formation of the beta-sheet structure of Abeta when undergoing peptide aggregation is important for Abeta's membrane perturbing properties. This effect ca...
متن کاملElectron capture dissociation mass spectrometric analysis of lysine-phosphorylated peptides
Phosphorylation of proteins is an essential signalling mechanism in eukaryotic and prokaryotic cells. Although N-phosphorylation of basic amino acid is known for its importance in biological systems, it is still poorly explored in terms of products and mechanisms. In the present study, two MS fragmentation methods, ECD (electron-capture dissociation) and CID (collision-induced dissociation), we...
متن کاملThioflavine T interaction with synthetic Alzheimer's disease beta-amyloid peptides: detection of amyloid aggregation in solution.
Thioflavine T (ThT) associates rapidly with aggregated fibrils of the synthetic beta/A4-derived peptides beta(1-28) and beta(1-40), giving rise to a new excitation (ex) (absorption) maximum at 450 nm and enhanced emission (em) at 482 nm, as opposed to the 385 nm (ex) and 445 nm (em) of the free dye. This change is dependent on the aggregated state as monomeric or dimeric peptides do not react, ...
متن کاملMass spectrometric survey of peptides in cephalopods with an emphasis on the FMRFamide-related peptides.
A matrix-assisted laser desorption/ionization (MALDI) mass spectrometric (MS) survey of the major peptides in the stellar, fin and pallial nerves and the posterior chromatophore lobe of the cephalopods Sepia officinalis, Loligo opalescens and Dosidicus gigas has been performed. Although a large number of putative peptides are distinct among the three species, several molecular masses are conser...
